Nació el 18 de agosto de 1922 en Saint Louis, aunque creció en Brooklyn.

Vendedora y modelo antes de iniciarse en Broadway en el año 1941.

En el cine participó en más de un centenar de películas y destacó en Doble vida (George Cukor, 1948).

Posteriormente aparece en Una vida marcada (R. Siodmak, id.), The Great Gatsby (Elliott Nugent, 1949), Winchester 73 (Anthony  Mann, 1950), Yo amé a un asesino (John Berry, 1951), Un lugar en el sol (G. Stevens, id.), La torre de los ambiciosos (Robert Wise, 1954), I Died a Thousand Times (S. Heisler, 1955), La noche del cazador (Charles Laughton, id.), The Big Knife (Robert Aldrich, id.), El diario de Ana Frank (Stevens, 1959), Lolita (Stanley Kubrick, 1962), Confidencias de mujer (G. Cukor, id.), Un retazo de azul (G. Green, 1965), Alfie (L. Gilbert, 1966), Harper investigador privado (J. Smight, id.), El presidente (Wild in the Streets, B. Shear, 1968), Mamá sangrienta (P. Obtuvo el Oscar como mejor actriz de reparto por sus trabajos en El diario de Ana Frank (1959) y en Un retazo de azul (1965),

Escribió varias obras de teatro y publicó en 1980 su autobiografía: Shelley Also Known As Shirley.

Estuvo casada con Paul Meyer, su primer marido, después contrajo matrimonio con el actor italiano Vittorio Gassman, con quien tuvo a su hija Vittoria-Gina. Su tercer marido fue el actor Tony Franciosa, del que se divorció a los tres años de casarse.

Shelley Winters murió el 14 de enero de 2005 por deficiencia cardíaca en el Centro de Rehabilitación de Beverly Hills. La actriz había sido hospitalizada en octubre después de sufrir un ataque cardíaco.
