Entra en contacto con el teatro en el colegio, participando en la gran mayoría de las representaciones escolares.
Cursó estudios en la universidad de esta ciudad con el objetivo de seguir los pasos de su padre en la escuela diplomática.
Cambió el rumbo de su vida cuando se hizo granjero en Canadá, aunque abandonó pronto para dedicarse a la interpretación.
Durante mucho tiempo viajó con distintas compañías ambulantes e interpretó un pequeño papel en la película Su majestad el americano (1919). Trabajó con regularidad durante el resto del periodo mudo pero pasó desapercibido hasta que encarnó al verdugo de un chivato, en Código penal (1931) de Howard Hawks.
El doctor Frankenstein (1931) dirigida por James Whale, fue una película que en principio interpretaría Bela Lugosi, como continuación de su mítico Drácula (1931), aunque el enfrentamiento del actor con la productora Universal les llevó a sustituirlo por Karloff, que resultaba más patético que terrorífico como monstruo. La momia de Karl Freund, El caserón de las sombras de James Whale, y La máscara de Fu-Manchú de Charles Brabin (todas ellas de 1932), le convirtieron en el maestro del cine de terror. Cuando la moda de las películas de terror pasó, se dedicó a papeles de reparto y a protagonizar películas de serie B.
Actuó como estrella para una película de primera clase, en The climax (1944), con la que la Universal pretendía repetir el éxito de Claude Rains en la versión de 1943 de El fantasma de la ópera. Sus últimas películas fueron a menudo de bajo presupuesto, con la excepción del filme de Peter Bogdanovich El asesino anda suelto (1967).
Boris Karloff falleció de neumonía en el King Edward VII Hospital, Midhurst, Sussex, el 2 de febrero de 1969.
