Nació el 10 de febrero de 1930 en Detroit (Estados Unidos). Se trasladó a principios de los años 50 a Los Angeles para convertirse en actor.

Ingresó en la Fox a principios de los cincuenta. Fue el jovencito enamorado de una chica de buena familia a bordo del más famoso y trágico trasatlántico de la historia, el Titanic, en la película dirigida por Jean Negulesco (1953). Obtuvo gran fama al interpretar al protagonista de La verdadera historia de Jesse James (1957), y por su aparición en La pantera rosa (Blake Edwards, 1964).

Otros de sus trabajos son: Situación desesperada (L. Milestone, 1951), Beneath the 12 Mile Reef(R. D. Webb, 1953), El príncipe valiente (Henry Hathaway, 1954); Lanza rota (Edward Dmytryk, id.),Los jóvenes caníbales (All the Fine Young Cannibals, M. Anderson, 1960) I sequestrati di Altona (Vittorio De Sica, 1962) y Harper, investigador privado (J. Smight, 1966).

Conoció a la actriz Natalie Wood cuando tenía 26 años y ella sólo 18, y desde entonces formaron una de las parejas más famosas de Hollywood. Se casaron en diciembre de 1957. Después se casaría con una vieja amiga, Marion Marshall, en 1963 y tuvo una hija, aunque la relación no duró mucho tiempo. En 1971, Wagner y Wood se reencontraron en un restaurante y se dieron cuenta que seguían enamorados. Ella abandonó a su marido y se volvieron a casar el 16 de junio de 1972 a bordo del yate Splendour. En 1981, Natalie cayó desde la cubierta del mismo yate ahogándose. Diez años después, volvió a casarse, con la también actriz Jill St. John.
