Nació el 26 de febrero de 1928 en Nueva Orleans (Estados Unidos).
Se crió en el seno de una familia de origen criollo francés, que fue su primera lengua. Su padre fue un reconocido violinista. Aprendió a tocar el piano de su cuñado, el guitarrista de jazz Harrison Verrett.
Fue apodado como Fats (Gordo), porque pesaba más de 100 kilos.
Pianista con influencias del boogie-woogie, es descubierto en 1948 en el Hideway Club, por Lew Chudd, de Imperial Records. Allí se encontró con Dave Bartholomew, hombre clave en la música de los años 50, y juntos compusieron su primer tema,The fat man. Su primer álbum, Carry on Rockin', se lanzó en noviembre de 1956. Consiguió 17 discos de oro, cifra superada únicamente por Beatles, Elvis Presley y Bing Crosby.
Entre sus canciones destacan: Son Goin' home en 1952; You said you loved me, Please don't leave me y Going to the river, en 1953; Love me, en 1954; All by myself, I can't go on y Ain't that a shame, en 1955; I'm in love again, Bo weevil, Blue monday y Blueberry Hill en 1956; It's you love, I'm walkin' y I still love you, en 1957; y Whole lotta lovin' en 1958.
Desde 1963, año en que Imperial dejó de existir como editora independiente edita a través del sello ABC de Nashville, siendo su más destacado tema la versión del Lady Madonna de los Beatles en 1968.
El 1 de septiembre de 2005, el legendario músico fue dado por desaparecido por familiares y amigos incapaces de contactar con el músico tras el azote del huracán "Katrina" en Nueva Orleans donde se encontraba. Fue rescatado un día después cuando su hija, Karen Domino White, lo identificó en una fotografía de un diario en la que se veía a un hombre siendo ayudado a salir de un bote por autoridades.
Antoine Fats Domino falleció el miércoles 25 de octubre de 2017 en Nueva Orleans, después de una larga enfermedad.
